Bcl-2、CytC和caspase-3在实验性精索静脉曲张大鼠生精细胞中的表达及意义(1)

     [摘要] 目的 探讨实验性精索静脉曲张(EV)青春期大鼠生精细胞Bcl-2、细胞色素C(CytC)和caspase-3的表达及意义。 方法 青春期雄性Wistar大鼠72只,随机分为6组,每组12只,分别为A1组、A2组、A3组、B1组、B2组、B3组,A1组、A2组和A3组为实验组,B1组、B2组和B3组为对照组。实验组大鼠通过行左肾静脉部分结扎术制作左侧精索静脉曲张模型(ELV);对照组只显露和游离左肾静脉,不结扎,其余步骤与实验组相同。模型建立后2、4、8周分别处死A1和B1组、A2和 B2组、A3和B3组大鼠,取左侧睾丸,并用免疫组化方法检测左侧睾丸生精细胞Bcl-2、CytC和caspase-3的表达,用TUNEL法检测左侧睾丸生精细胞的凋亡并计算凋亡指数(AI)。 结果 实验组大鼠Bcl-2表达较对照组显著降低(P < 0.05),而实验组CytC、caspase-3表达较对照组明显升高(P < 0.05),实验组AI较对照组明显升高(P < 0.05)。 结论 精索静脉曲张大鼠生精细胞Bcl-2表达降低,CytC和caspase-3表达升高,从而导致睾丸生精细胞凋亡增加。

    The effect of experimental left varicocele on Bcl-2, CytC and caspase-3 in spermatogenic cells of adolescent rats

    [Abstract] Objective To study the effect of experimental left varicocele on Bcl-2, CytC and caspase-3 in spermatogenic cells of adolescent rats. Methods 72 male Wistar rats were divided into 6 groups randomly, each group had 12 rats, respectively A1 group, A2 group, A3 group, B1 group, B2 group and B3 group. A1, A2 and A3 group were the experimental groups, B1, B2 and B3 groups were the control groups. The left renal vein of experimental groups were partial ligated. The left renal vein of control groups were only revealed and no ligature, the remaining steps were the same as the experimental groups. 2, 4 and 8 weeks after surgery, the testes of the ELV models and controls were extirpated. The expression of Bcl-2, CytC and caspase-3 were studied by immunohistochemistry. The apoptosis index was detected by TUNEL. Results There was a remarkable decline of the expression of Bcl-2 of the experimental groups. There was a remarkable increase of AI and the expression of CytC and caspase-3 of the experimental groups (P < 0.05). Conclusion Varicocele can reduce the expression of Bcl-2, increase the expression of CytC and caspase-3 and the apoptosis of spermatogenesis cells ......
