[摘要] 目的 探讨大蒜素对腹主动脉缩窄大鼠左室肥厚及钙调神经磷酸酶信号通路影响。 方法 行腹主动脉部分缩窄术制作压力负荷左室肥厚模型,40只大鼠随机分为假手术组、模型组、大蒜素组和缬沙坦组,每组10只。饲养6周后测量体重(BW),通过测量左室重量(LVW)及计算左室肥厚指数(LVMI),行病理切片以证明心肌肥厚。检测左室血流动力学参数及血浆内血管紧张素Ⅱ(AngⅡ)、内皮素1(ET1)水平,Western Blot法检测心肌钙调神经磷酸酶(CaN-A,其活性亚基)、C-fos蛋白的表达。 结果 与假手术组相比较,各手术组(模型组、大蒜素组和缬沙坦组)LVW及LVMI明显增加(P < 0.05),病理切片示心肌细胞肥大,组织结构紊乱,各手术组血浆内AngⅡ、ET1明显增高(P < 0.05),其中缬沙坦组血管紧张素Ⅱ(AngⅡ)水平升高最为明显(P < 0.01)。与模型组相比较,大蒜素组、缬沙坦组心肌CaN、C-fos蛋白表达明显受抑制(P < 0.05)。 结论 大蒜素对延缓腹主动脉缩窄所致的心肌肥厚有一定的作用,其机制可能是减少对CaN信号通路的激活。

Effects of garlcin on cardiac herpertrophy in rats with abodominal aorta coarctation and the mechanism of calcineurin signaling pathways

[Abstract] Objective To investigate the effects of garlicin on left ventricular hypertrophy (LVH) in rats with abdominal aorta coarctation (AAC) and calcineurin signaling pathways. Methods The left ventricular hypertrophy model was induced by AAC. Forty female spraque dawley (SD) rats were randomly divided into four groups: sham-operated group, AAC model group, garlicin group and Valsartan group. After six weeks, the body weight (BW) were observed,then the LVH was evidenced by calulating the elevated left ventricular weight (LVW), left ventricular mass index (LVMI) and histopathology. Hemodynamic parameters of left ventricular and the concertration of angiotension Ⅱ(AngⅡ) and endothelia 1 (ET1) in plasma were determined. The expressions of CaN-A (the catalytic subumit of calcineurin) and C-fos were determined at the protein levels by Western Blot.
