氟伐他汀对同型半胱氨酸诱导的凝集素样氧化低密度脂蛋白受体—1表达的影响(1)
[摘要] 目的 探讨氟伐他汀对同型半胱氨酸(Hcy)诱导的人脐静脉内皮细胞(HUVECs)凝集素样氧化低密度脂蛋白受体-1(LOX-1)信使核糖核酸(mRNA)表达的影响。 方法 分离培养HUVECs,分为正常对照组、Hcy刺激组:100 μmol/L Hcy与细胞孵育24 h,3种不同浓度的氟伐他汀干预组:分别以不同浓度的氟伐他汀(0.1、1、10 μmol/L)预孵育细胞24 h,再与100 μmol/L Hcy共同培养24 h,应用逆转录-聚合酶链反应(RT-PCR)检测LOX-1 mRNA的表达。 结果 Hcy显著诱导HUVECs LOX-1 mRNA的表达,氟伐他汀抑制Hcy诱导的LOX-1 mRNA的表达,并呈现一定剂量依赖性。 结论 Hcy可能通过LOX-1介导其内皮损伤作用,氟伐他汀可能通过减少LOX-1过度表达发挥其抗动脉粥样硬化作用。

[Abstract] Objective To investigate the effect of fluvastatin on the expression of LOX-1 induced by homocysteine (Hcy) in cultured human umbilical vein endothelial cells (HUVECs). Methods Human umbilical vein endothelial cells were cultured and treated with Hcy (100 μmol/L for 24 hours) alone, or pre-treated with fluvastatin (0.1, 1 or 10 μmol/L for 24 h), followed by Hcy treatment. After respective treatment, the expression of lectin-like ox-LDL receptor-1 (LOX-1) was measured by reverse transcription-polymerase chain reaction (RT-PCR). Results Hcy (100 umol/L) treatment up regulated the expression of LOX-1 in HUVECs. Pretreatment of HUVECs with fluvastatin (0.1, 1 or 10 μmol/L) decrease Hcy-induced expression of LOX-1. A high concentration of fluvastatin (10 umol/L) was more potent than the lower concentration (1 μmol/L). Conclusion These observations indicate that endothelial dysfunction elicited by Hcymay beassociated with an increase of LOX-1 expression. Inhibition of expression of LOX-1 may be another mechanism of beneficial effects of statins in vascular diseases.

动脉粥样硬化是心脑血管疾病的病理基础,内皮功能障碍则是动脉粥样硬化的始动环节。研究证实同型半胱氨酸(Hcy)能够导致内皮损伤,促进动脉粥样硬化形成[1]。已知他汀类药物具有抗动脉粥样硬化作用,其中独立于降脂作用以外的内皮保护功能尤为重要。LOX-1作为氧化型低密度脂蛋白(oxLDL)的内皮细胞受体,通过介导oxLDL在内皮细胞的多种生物学效应,参与动脉粥样硬化的形成[2]。多种炎症因子、氧化应激因素都可诱导LOX-1的表达。但目前鲜有关于Hcy对内皮细胞LOX-1表达诱导以及氟伐他汀对内皮细胞LOX-1表达影响的研究。故本研究拟探讨氟伐他汀在体外对Hcy诱导的LOX-1表达所具有的影响。
1 材料与方法
1.1 一般材料
DL-同型半胱氨酸(Sigma公司),氟伐他汀原药粉(Novartis公司,批号:X0362),RPMI1640培养基、胎牛血清、人AB血清(GibcoBrl公司),青霉素、链霉素、胶原酶Ⅰ型(Sigma公司),0.25%胰蛋白酶及0.02%EDTA(HyClone公司),HEPES、PBS缓冲液、内皮生长因子(Sigma公司),肝素钠(Solarbio公司),内皮细胞鉴定试剂盒(福州迈新生物技术开发有限公司),逆转录试剂盒(Promega公司),通用PCR优化试剂盒Ⅰ(博大泰克公司)。
1.2 人脐静脉内皮细胞分离培养与鉴定
无菌条件下取健康产妇脐带,PBS缓冲液反复冲洗后,加0.1%胶原酶,37℃消化15 min,离心后加完全培养基RPMI1640(10%胎牛血清,10%人AB血清,100 U/mL青霉素,100 μg/mL链霉素,50 ng/mL内皮生长因子,50 μg/mL肝素钠储存液),置37℃、5%CO2孵箱中培养,24 h后换液,待人脐静脉内皮细胞80%融合,用胰蛋白酶-EDTA消化传代,经Ⅷ因子鉴定后,第2~3代用于实验。 (赵颖 赵兴山 孙华毅 胡文兰)
