瑶药扶芳藤对血管内皮细胞缺氧损伤的保护作用(1)
[摘要] 目的 研究瑶药扶芳藤乙醇、正丁醇提取物对血管内皮细胞ECV-304缺氧损伤的作用影响。 方法 体外培养ECV-304细胞,MTT法测定扶芳藤提取物细胞毒性,以Na2S2O4诱导ECV-304细胞缺氧损伤模型,分别将扶芳藤乙醇、正丁醇提取物作用于细胞24 h后,MTT法检测细胞存活率;收集细胞上清液,检测LDH露出率;台盼蓝计数检测细胞死亡率。 结果 扶芳藤乙醇和正丁醇提取物浓度为25~400 μg/mL,对ECV-304细胞均无细胞毒作用;与模型对照组比较,扶芳藤乙醇(浓度50 μg/mL)、正丁醇(浓度50、100、200 μg/mL)提取物对缺氧损伤ECV-304细胞存活率有显著提高,差异有统计学意义(P < 0.05或P < 0.01)。与模型对照组比较,扶芳藤正丁醇提取物(浓度100 μg/mL)能明显降低缺氧损伤ECV-304细胞上清LDH露出率,(P < 0.05);与模型对照组比较,扶芳藤正丁醇提取物(浓度50、100、200 μg/L)能抑制缺氧损伤细胞ECV-304凋亡(P < 0.01)。 结论 扶芳藤提取物对Na2S2O4诱导的血管内皮细胞缺氧损伤有一定保护作用。

[Abstract] Objective To investigate the effect of ethanol and butanol extract of Yao medicine Euonymus fortunei on ECV-304 hypoxia injury of vascular endothelial cells. Methods ECV-304 cells were cultured in vitro, and the cytotoxicity of Euonymus fortunei extract was determined by MTT. ECV-304 cells hypoxia injury model was induced by Na2S2O4, respectively Euonymus fortunei ethanol and n-butanol extracts from cells after 24 h, the survival rate of cells was detected by MTT. The cell supernatant was collected for detection of LDH, the cell apoptosis rate was detected by trypan blue staining. Results Euonymus fortunei ethanol and n-butanol extract was in 25-400 μg/mL concentration range, which was no cytotoxic effect on ECV-304 cells. Compared with the model group, the ECV-304 cells survival rate of Euonymus fortunei (ethanol concentration of 50 μg/mL), n-butanol (concentration of 50, 100, 200 μg/mL) extract on hypoxia injury was significantly increased (P < 0.05 or P < 0.01). Compared with the model group, the ECV-304 cells LDH exposed rate of Euonymus fortunei n-butanol extract (concentration of 100 μg/mL) was significantly decreased (P < 0.05). Compared with the model group, the ECV-304 cells apoptosis rate of Euonymus fortunei n-butanol extract (concentration of 50, 100 and 200 μg/mL) was inhibited (P < 0.01). Conclusion The extract of Euonymus fortunei has protective effect on Na2S2O4 induced hypoxia injury of vascular endothelial cells.

血管內皮细胞是连续被覆在全身血管内膜的一层细胞群,其在血管生成、止血、创伤修复等病理和生理过程中起着重要作用,血管内皮细胞的损伤与多种心血管疾病(如动脉粥样硬化、高血压、血栓形成等)有密切的关系,从整体、细胞、分子水平研究对血管内皮细胞具有保护作用的药物,对心血管疾病的防治研究具有重要意义。现代研究表明,活血化瘀类中药及其有效成分能从多个方面保护血管内皮细胞免受损伤和促进内皮细胞增殖,从而为中药治疗血管内皮细胞功能受损相关性疾病提供了药理学依据[1-5]。
