柚皮素对人肺鳞癌NCI—H2170细胞增殖、凋亡的影响(1)
[摘要] 目的 探討柚皮素对人肺鳞癌细胞NCI-H2170的增殖、凋亡及细胞周期的影响。 方法 用MTT法检测柚皮素对NCI-H2170细胞增殖的影响,Hoechst33258/PI染色检测NCI-H2170细胞形态学变化,流式细胞术(FCM)检测细胞周期的改变。Western blot检测柚皮素处理NCI-H2170细胞24 h后PARP、Bcl-2、Bid、procaspase-3及procaspase-8蛋白的表达。 结果 柚皮素处理人肺鳞癌NCI-H2170细胞24 h后,浓度依赖性地抑制NCI-H2170细胞增殖;其中12.5 μmol/L处理组与对照组比较,差异无统计学(P > 0.05);25、50、100 μmol/L处理组与对照组比较,差异有统计学意义(P < 0.05)。流式细胞仪分析结果显示:不同浓度的柚皮素处理NCI-H2170细胞24 h后,与对照组比较,12.5 μmol/L处理组差异无统计学意义(P > 0.05);25、50、100 μmol/L处理组G1期细胞显著增多,S期及G2期细胞显著减少,差异均有统计学意义(P < 0.05)。25、50、100 μmol/L处理组与对照组相比,柚皮素上调Bid表达,促进PARP降解,Bcl-2、procaspase-3和procaspase-8降低,差异均有统计学意义(P < 0.05)。 结论 柚皮素抑制人肺鳞癌NCI-H2170细胞增殖,并能诱导NCI-H2170细胞产生G1期阻滞,诱导细胞凋亡。

[Abstract] Objective To investigate the effects of Naringenin on the proliferation, apoptosis and cell cycle of human lung squamous cell carcinoma cell line NCI-H2170. Methods The proliferation of NCI-H2170 cells was detected by MTT assay. The morphological changes of NCI-H2170 cells were detected by Hoechst33258/PI staining. Cell cycle changes were measured by flow cytometry (FCM). Western blot was used to detect the expression of PARP, Bid, Bcl-2, procaspase-3 and procaspase-8 in NCI-H2170 cells treated with naringenin. Results The proliferation of human lung squamous cell carcinoma NCI-H2170 cells were inhibited in a dose-dependent manner after treating with Naringenin for 24 h. It was not significant difference between 12.5 μmol/L treatment group and the control group (P > 0.05).However, 25, 50, 100 μmol/L treatment groups were significantly different comparing to the control group (P < 0.05). The results of flow cytometry analysis showed that it was not significant difference between 12.5 μmol/L treatment group and the control group (P > 0.05). However, after NCI-H2170 cells were treated with 25, 50, 100 μmol/L naringenins, the number of cells had significantly increased in G1 phase, desceased in S phase and G2 phase, comparing to the control group (P < 0.05). Naringenin up-regulated Bid expression, promoted the degradation of PARP and the decrease of Bcl-2, proaspase-3 and proaspase-8 in NCI-H2170 cells, comparing to the control group (P < 0.05). Conclusion Naringenin inhibits the proliferation of human lung squamous cell carcinoma cell line NCI-H2170 and induces the G1 phase arrest in NCI-H2170 cells and induces cell apoptosis., 百拇医药(曾德贵 陈保安 吴尚 黄慧 伍俊)
