HPLC法测定冬虫夏草胶囊中腺苷的含量(1)
摘要 目的:建立院内制剂冬虫夏草胶囊中腺苷含量的测定方法,为提高该制剂的质量标准提供依据。方法:采用高效液相色谱法,色谱柱为Hibar-C18柱(4.6 mm×150 mm,5 μm);流动相为pH值6.5的磷酸盐缓冲液-甲醇(85∶15);检测波长为260 nm;柱温30 ℃;流速为1.0 mL/min;进样量为10 μL。结果:腺苷在0.032 7~0.163 5 μg范围内,线性关系良好(R2=0.999 4);加样回收率为99.8%~101.5%;测定3个批次的冬虫夏草胶囊中腺苷的平均含量分别为0.91 mg/粒,0.92 mg/粒,0.93 mg/粒,相对标准偏差(RSD)分别为0.44%、1.15%、2.06%。结论:该方法简便,准确,重现性好,灵敏度高,可作为冬虫夏草胶囊质量控制指标;所测3批冬虫夏草胶囊中腺苷的含量相近,质量可靠,可用于临床治疗。

Determination of Adenosine Content in Dongxia Chongcao Capsules by HPLC

Abstract Objective:To develop an HPLC method for the determination of adenosine in Dongxia Chongcao Capsules and provide a basis for improving the quality standards of the preparation. Methods:HPLC was used and the analytical column was Hibar-C18 (4.6 mm×150 mm,5 μm). The mobile phase was phosphate buffer pH6.5-methanol (85∶15). The detection wavelength was 260 nm. The column temperature was 30 ℃. The flow was 1.0 mL/min. The injection volume is 10 μl. Results:Adenosine had a good linear relationship (R2=0.999 4) in the 0.032 7~0.163 5 μg range,and the sample recovery was 99.8%~101.5%. The average contents of adenosine in 3 batch slices of Chinese caterpillar fungus Capsules was 0.91 mg,0.92 mg,0.93 mg. The relative standard deviations (RSD) were 0.44%,1.15%,and 2.06%,respectively. Conclusion:The contents of adenosine in the 3 batches of Dongxia Chongcao Capsules are analogical. This method is simple,accurate and available for the quality control of Dongxia Chongcao Capsules. The 3 batches of Dongxia Chongcao Capsules have similar adenosine content and reliable quality,which can be used for clinical treatment.

冬虫夏草是麦角菌科真菌冬虫夏草菌Cordyceps sinensis(BerK.)Sacc.寄生在蝙蝠蛾科昆虫幼虫上的子座及幼虫尸体的复合体。作为我国特有的名贵药用真菌,其具有补肺益肾、止血化痰的功效。本院院内制剂虫草胶囊采用超微破壁技术,在避免了溶剂提取造成的活性成分的损失的同时,能够最大限度释放冬虫夏草的活性成分,便于机体的吸收利用。研究表明,冬虫夏草的活性成分主要包括:核苷、多糖、蛋白质、甾醇等,其中核苷类化合物是近年国内外研究的热点。核苷类化合物包括:腺苷、虫草素、鸟苷、肌苷等。腺苷是腺嘌呤核苷酸的简称,具有改善心脑血液循环、抑制中枢神经递质释放等活性,已被中华人民共和国药典作为冬虫夏草及其制品的质控指标[1]。因此,本实验采用了中华人民共和国药典的标准测定冬虫夏草胶囊中腺苷的含量,从而控制其质量。
目前针对腺苷含量的常见测定方法有薄层色谱法[2]、高效液相色谱法[3-9]和高效液相-质谱法[10]等。薄层色谱法较简单,但准确度与专属性不如另外2种方法;高效液相-质谱法准确度、精密度、专属性等性能方面优越,但其价格高昂对人员素质要求也较高,不利于方法的普及。高效液相色谱法分离效率高,选择性好,检测灵敏度高,操作自动化,有利于该方法在规模化批量检测中的运用。, http://www.100md.com(许多 刘梅 杨波 刘 静 刘子修 司梁宏 陆瑜)
