过氧化物酶增殖物激活受体—γ和核因子E2相关因子—2在肾脏衰老中的作用

叶婷+宁勇[摘要] 目的 探讨过氧化物酶增殖物激活受体-γ(PPARγ)和核因子E2相关因子-2(Nrf2)在肾脏衰老中的作用。方法 以3个月龄和24个月龄大鼠作为青年组和衰老组(n = 10),PAS染色观察肾脏组织病理改变,肾脏组织匀浆测定H2O2、丙二醛(MDA)含量,Western Blot法检测肾脏组织髓过氧化物酶(MPO)、PPARγ和Nrf2蛋白质表达。 结果 衰老组大鼠肾脏组织MDA[(44.42±4.17)μmol/mg比(26.23±1.43)μmol/mg]、H2O2[(239.90±39.38) μmol/μg比(110.00±19.49)μmol/μg]含量较青年组均明显增加,差异有统计学意义(P 0.05)。 结论 氧化损伤在肾脏衰老中发挥重要作用,可能是通过PPARγ表达下降介导。
