目的 探讨三七总皂苷(PNS)对子宫内膜癌Ishikawa和HEC-1A细胞增殖、侵袭和凋亡的影响及其作用机制。方法 应用不同浓度的PNS分别作用于Ishikawa和HEC-1A细胞24、48、72 h,采用MTT法检测细胞增殖率;Transwell检测细胞侵袭能力;流式细胞仪检测细胞凋亡水平;采用RT-PCR和Western blot分别检测VEGF、PI3K、AKT的mRNA和蛋白表达水平。 结果PNS可以显著抑制Ishikawa和HEC-1A细胞的增殖。与control组比较,PNS处理的Ishikawa和HEC-1A细胞侵袭显著下降(P<0.05),细胞凋亡率明显升高(P<0.05),VEGF和PI3K的表达水平下降(P<0.05)。结论PNS能够抑制Ishikawa和HEC-1A细胞的增殖和侵袭,诱导细胞凋亡,其过程可能与VEGF/PI3K/AKT信号通路的抑制有关。
