基于网络药理学的补阳还五汤防治阿尔茨海默病的机制研究

    [摘要] 目的 预测发现补阳还五汤防治阿尔茨海默病(AD)的作用靶标,探讨多成分-多靶点-多通路的协同作用机制。 方法 基于网络药理学方法针对补阳还五汤的方剂组成,采用多元网络药理学方法预测补阳还五汤的体内作用靶标,通过BATMAN-TCM等数据库对比补阳还五汤抗AD药物靶标,最终采用Cytoscape软件构建补阳还五汤成分-靶点-通路的网络。 结果 补阳还五汤中活性成分涉及多个与AD直接相关的体内靶标,如载脂蛋白E(APOE)等。同时涉及多个代谢通路,如甘油磷脂代谢、胆碱能突触、胆固醇代谢、AD通路等。 结论 补阳还五汤主要通过多成分协同起效,如1-十六碳烯对载脂蛋白E调节间接起到调控疏水性淀粉样β(Aβ)沉淀形成、藏花醛对乙酰胆碱酯酶和丁酰胆碱酯酶的抑制作用等,进而延缓AD的发生发展。

    [Abstract] Objective To predict and discover the target of Buyang Huanwu Decoction in the prevention and treatment of Alzheimer′s disease (AD) and explore the synergistic mechanism of multi-component, multi-target and multi-pathway. Methods According to the composition of Buyang Huanwu Decoction, the in vivo target of Buyang Huanwu Decoction was predicted by multiple network pharmacology methods. The target of anti-AD drug was selected by BATMAN-TCM and other databases. Finally, the components of Buyang Huanwu Decoction were constructed by Cycaptose software for target path network.
