    [摘要] 载脂蛋白E(ApoE)是一种有明显基因多态性、多功能蛋白，与脂蛋白的代谢有密切联系，其缺失和变异都会引起体内代谢失常及功能紊乱，甚至引起一系列疾病和并发症。近年来，ApoE-/-小鼠主要应用于动脉粥样硬化、糖尿病、神经退化性疾病、肾脏病等。为了更好地研究ApoE在各疾病模型中的应用机制，为临床疾病治疗提供更加有力的依据，本文总结了人类ApoE基因型与ApoE蛋白相关的几种疾病的发生机制，并对体外研究疾病发展过程的动物模型进行了综述。
