乳腺癌中转化生长因子TGFβ1mRNA及蛋白表达及意义(1)
【摘要】 目的 探讨转化生长因子TGFβ1 mRNA及其蛋白在乳腺癌中的表达及其意义。方法 采用RT-PCR和免疫组织化学法分别检测96例乳腺癌新鲜组织及对应60例石蜡组织中TGFβ1 mRNA及蛋白的表达。结果 乳腺癌中TGFβ1 mRNA及蛋白的高表达率分别为45.83%、43.33%,明显高于其在乳腺良性增生组织(26.67%,20%)及正常乳腺组织(9.09%,0)中的表达,差异有统计学意义(P1 mRNA及其蛋白高表达率分别为64.29%(36/56)、57.14%(20/35),均明显高于腋淋巴结无转移组的20.00%(8/40)、24.00%(6/25),两组表达的差异有统计学意义(P1 mRNA及其蛋白在乳腺癌发病发展过程中起重要促进作用,提示TGFβ1基因可能是乳腺癌的重要促淋巴转移因子,有望成为治疗的靶标及有价值的预后判断指标。

Expression and significance of the transforming growth factor β1 in breast cancer

【Abstract】 Objective To investigate the expression of transforming growth factor β1(TGFβ1)in breast cancer and its significance.MethodsTGFβ1 mRNA was detected in 96 fresh tumor tissues of breast cancer using semi-quantitative reverse transcription polymerase chain reaction(RT-PCR),and TGFβ1 protein was examined in 60 corresponding formalin-fixed paraffin-embedded tissues by immunohistochemistry.Results The overexpression rates of both TGFβ1 mRNA and protein in breast cancer tissues were 45.83%,43.33%,respectively,which were higher than that in innocence hyperplastic tissues of breast(26.67%,20%)and normal breast tissues(26.67%,20%)with significant difference(P1 mRNA and protein overexpression rates were 64.29%(36/56),57.14%(20/35)and 20.00%(8/40),24.00%(6/25),respectively,the differentence of which was statistically significant(P1 mRNA and protein may play an important role in tumorigenesis and progression of breast cancer,and TGFβ1 gene may be one of factors accelerating lymph node metastasis of breast cancer,which may become a target of treatment and a prognostic biomarker in breast cancer.

1 材料与方法
1.1 临床资料 96例乳腺癌新鲜组织样本取自济南市历下区医院及山东大学第二医院2002~2005年手术患者,其中60例对应石蜡标本取自该两院病理科存档蜡块,患者均为女性,年龄30~87岁,中位年龄57岁。术前均未接受化疗和/或放疗。所有患者均经术后病理检查证实,按WHO最新标准对各患者进行病理学分级和分型[6],其中浸润性导管癌71例,其他类型癌25例;临床分期按国际抗癌联盟(UICC)TMN分期[7]:I~Ⅱ期64例,Ⅲ+Ⅳ期32例;腋窝淋巴结转移56例,无转移40例(石蜡标本资料另记)。选取同期乳腺良性增生组织15例及正常乳腺组织11例。
1.2 主要试剂及仪器 Trizol试剂盒及SuperScriptTMⅡReverseTranscriptase试剂盒为美国Invitrogen公司产品。美国MJ PTC-220多通道PCR仪;美国LAMBDA BIO 20双光束紫外分光光度计;美国BIO-RAD GD Doc2000凝胶电泳图像分析系统。TGFβ1(1:200)蛋白为兔抗人多克隆抗体,美国Santa Cruz公司产品。即用型SP检测试剂盒购自北京中杉金桥公司(美国Zymed公司产品)。, 百拇医药(赵国仁 朱凌冬 安 纲)
