毛细管气相色谱法测定糜蛋白酶中残留有机溶剂
【摘要】 目的 建立测定糜蛋白酶中有机溶剂丙酮残留量的方法。方法 采用毛细管气相色谱法,色谱柱为Supelco 25301-U型石英毛细管柱,检测器为氢火焰离子化检测器,载气为高纯度氮气,水为溶解介质,检测了糜蛋白酶中丙酮的残留量。结果 丙酮在样品中达到完全分离,在1.59~25.50 ppm浓度范围内具有良好的线性,线性方程为A=306.0C+184.3,r=0.999 5,检测限为0.03 ppm,方法回收率为99.9%,RSD 为1.7%。结论 本法能快速准确的检测糜蛋白酶中有机溶剂丙酮残留量,可用于糜蛋白酶质量控制中的有机残留溶剂检查。
