【摘要】 目的 探讨三磷酸腺苷(ATP)在大鼠胰岛分离过程中抑制胰酶对胰岛细胞的消化作用。方法 将20只SD大鼠平均分为两组，处理组在胰岛分离全过程中加用ATP进行干预，对照组则不加ATP处理，采用Ficoll非连续密度离心纯化后对两组胰岛细胞的凋亡、胰岛产量及活力方面进行比较性研究。结果 加用ATP的处理组的胰岛细胞凋亡率显著低于对照组(P<0.01),处理组胰岛产量和胰岛素释放水平均显著高于对照组(P<0.01)。结论 胰岛分离过程中加用一定浓度ATP对胰岛具有抑制凋亡和保护胰岛活力的作用。

    【Abstract】 Objective To observe the anti-apoptosis effect of ATP in rat islets isolation.Methods The rats were devided into 2 groups according whether with the treatment of ATP during the islets isolation.The purified islets by Ficoll were compared between the groups.Results The outcome and anmouts of insulin release under the glucose stimulating of isolated and purified islets in ATP group were superior to those in the control group(P<0.01).The number of apoptosis islet cells in ATP group were fewer than those in control group(P<0.01).Conclusion With the treatment of ATP during the islets isolation may protect the islets from trypsin digestion and prevent apoptosis of islet cells. ......
