pTAT-HA质粒载体在受体菌DH5α中的转化及鉴定
【摘要】 目的 探索pTAT-HA质粒在受体菌DH5α中转化的方法。方法 配制LB培养基,将pTAT-HA质粒转化至DH5α,培养筛选成功转化子,电泳鉴定。结果 pTAT-HA质粒转化菌可在Amp+LB培养基中生长,电泳结果显示质粒大小准确无误(约3000 bp)。结论 pTAT-HA质粒转化成功,转化菌可冷冻保存质粒,提取的质粒可用于下游分子生物学实验。

【Abstract】 Objective To explore the method thatDH5α was transformed with pTAT-HA vector.Methods LB medium was prepared,pTAT-XIAP vector was transformed into E.coli.DH5α.competent cells.Results Transformants were cultured on LB medium and selected.The plasmids were extracted,purified andidentified by agarose gel electrophoresis.Transformed DH5α Could grow on LB medium containing ampicillin,gel electrophoresis analysis showed that the molecular weight of pTAT-XIAP vector was about 3000 bp consistent withthe vector.Conclusion The pTAT-XIAP vector was succesfully transformed into the DH5α and the transformants were stored by cryopreservation,the purified plasmid could be used for molecular biologic experiments in the future.
