血红素加氧酶-1重组乳酸乳球菌灌胃对正常大鼠肠黏膜屏障的影响

【摘要】 目的 观察血红素加氧酶-1(HO-1)重组乳酸乳球菌同一剂量不同灌胃次数对正常大鼠肠黏膜屏障的影响。
方法 将40只健康雄性SD大鼠随机分为: HO-1重组乳酸乳球菌灌胃一次组(HO1t组,n=8)、HO-1重组乳酸乳球菌灌胃二次组(HO2t组,n=8)、HO-1重组乳酸乳球菌灌胃三次组(HO3t组,n=8)、PBS溶液灌胃一次组(PBS1t组,n=8)和乳酸乳球菌灌胃一次组(LL1t组,n=8)。各组灌胃剂量的容积均为1 ml,每次灌胃间隔24 h,在末次灌胃24 h后取回肠组织,检测肠组织Chiu’s 6级评分、髓过氧化物酶(MPO)活性,同时从右心室抽取血液行细菌学检查。
结果 各组肠黏膜Chius评分比较差异无统计学意义;与LL1t组比较,HO2t组和PBS1t组的肠组织内MPO活性较高,但比较没有统计学意义;HO2t组有1例、HO3t组有2例血培养阳性,其余各组血培养阴性。
结论 HO-1重组乳酸乳球菌对正常大鼠灌胃,不会随着灌胃次数增加而影响肠黏膜屏障的功能。

【Abstract】 Objective
To research the effect of lavaged with Lactococcus Lactis recombinant oxygenase-1 gene and heme on the rats intestine mucosa barrier as different times with the same dose. Methods 40 SD healthy male rats were randomly divided into one time lavaged with the LL1t Lactis recombinant HO-1 gene group(HO1t group, n=8), two times lavaged with the LL1t Lactis recombinant HO-1 gene group(HO2t group, n=8), three times lavaged with the LL1t Lactis recombinant HO-1 gene group(HO3t group, n=8), one time lavaged with pure phosptant buffer group(PBS1t group, n=8),one time lavaged with LL1t Lactis group(LL group,n=8).All rats were lavaged with 1 ml dose and 24 h between the times. The Chius grade and MPO activity in the low intestine, the blood in the right ventricle were detected.
Results It was no significantly different in Chius grade between the groups.Compared to the MPO activity of LL1t group, it was higher in HO2t group and HO3t group, but no significantly.1 in HO2t group and 2 cases in HO3t group were detected bacterial, other groups were not find.
Conclusion It is not effects the intestine mucosa barrier function following the lavaged dose of LL1t Lactis recombinant heme oxygenase-1 gene.
