多西苯醌联合塞来昔布对结肠癌细胞抑制作用的研究(1)

     【摘要】 目的 通过体外实验对比研究单独及联合应用多西苯醌(AA 861)与塞来昔布对结肠癌的抑制作用。方法 体外分组培养的结肠癌HT 29细胞,通过倒置相差显微镜观察AA 861与塞来昔布单独及联合应用48 h后细胞形态变化;通过细胞增殖试验(MTT)检测细胞吸光度,以此检测细胞增殖状态;应用免疫荧光染色方法在共聚焦显微镜下观察细胞内VEGF表达变化。结果 倒置相差显微镜观察到各药物组细胞形态发生明显变化,其中AA 861与塞来昔布联合应用组变化最明显;MTT结果提示两种药物联合应用组细胞增殖率最低;免疫荧光染色实验观察到细胞内VEGF表达减少,两种药物联合应用组VEGF表达量最低。结论 多西苯醌与塞来昔布联合应用比单独应用,更能有效抑制结肠癌,并且可能与抑制VEGF表达有关。

    【Abstract】 Objective To comparative investigate the influence of AA 861 alone or in combination with Celecoxib on the proliferation of colon carcinoma HT 29 cell line.Methods Group cultured colon cancer HT 29 cells,The morphological changes of HT 29 cells was observed by inverted phase contrast microscope after Docebenone(AA 861)and Celecoxib alone or in combination;By cell proliferation assay(MTT)detect cell absorbance so as to test cell proliferation;The expression change of VEGF was detected by Immunofluorescence staining experiments under Confocal microscope.Results We can observe that cell morphology chaned significantly under the inverted phase contrast microscope,and the group of AA 861and Celecoxib in combination changed most obvious; These results of MTT suggest that the group of two drugs in combination shows the lowest rate of cell proliferation.Immunofluorescence staining experiments revealed that the expression of VEGF was decreased,and the group of two drugs in combination has the lowest expression.Conclusion AA 861 and Celecoxib in combination can produce a superior anti colon cancer effection than AA 861 or Celecoxib alone,and this effect may be related with inhibit the expression of VEGF.
