温度与放置时间对产前筛查标本的影响(1)

     [摘要] 目的 探讨在产前筛查血清标本离心前放置时间及温度对游离人绒毛膜促性腺激素β亚基(free β-hcg)、甲胎蛋白(AFP)、游离雌三醇(uE3)测定结果的影响。 方法 额外收集产前筛查标本全血6 mL,分装至5支试管(水浴组加做一支用于对照),在室温(23℃~25℃)或水浴箱(35℃~37℃)下分别放置0.5 h、2 h、4 h、6 h、8 h后离心分离血清,以室温下0.5 h分离血清的标本作为对照组,各组与对照组分别进行比较分析。 结果 置室温组样本free β-hcg以约每小时0.9%的速率上升,AFP、uE3与对照组比较差异无统计学意义(P>0.05);水浴条件下free β-hcg以约每小时10.9%的速率迅速升高,AFP以每小时0.6%的速率上升,uE3则以每小时0.5%的速率降低。 结论 室温条件下,2 h内分离对结果影响较小,水浴条件对free β-hcg结果有严重影响,对其余两项也存在影响。产筛标本应尽量在较低温度下处理,建议不要对产前筛查标本进行37℃水浴处理。

    Effect of temperature and the duration on second-trimester serum markers

    [Abstract] Objective To examine the effect of the duration of storage of whole blood at different controlled temperatures on the concentrations of serum free-β-human chorionic gonadotropin (f-βhCG)、Alpha fetoprotein(AFP) and unconsecrated estriol(uE3). Methods We additionally collected 6 mL of whole blood of prenatal screeningspecimen, respectively put into 5 vacuum tubes(The water bath group should make an additional piece for comparison), placed at room temperature (23℃-25℃) or in the water bath box (35℃-37℃) for 0.5 h, 2 h, 4 h, 6 h, 8 h, after that, made a centrifugal separation to separate the blood serum, taken the specimen of blood serum separated atroom temperature for 0.5 h as the control group, and respectively compared and analyzed each group with the control group. Results In the room temperature group, f-βhCG had a rate of 0.9% per hour increasing. No significant difference was found in AFP and uE3(P>0.05). In the 37℃ group, f-βhCG had a rate of 10.9% per hour increasing rapidly. AFP had a rate of 0.6% per hour increasing, and uE3 had a rate of 0.5% per hour decreasing. Conclusion Under the condition of room temperature, to separate in 2 hours has a less influence on the result, and the water bath condition has a serious effect on the results of free β-hcg and the other two items. The prenatal screening specimen should be processed at a lower temperature as far as possible, and it is not suggested to make a water bath treatment of 37℃ for the prenatal screening specimen.
