心气阴两虚证心力衰竭大鼠血浆代谢组学研究(1)

     〔摘要〕 目的 研究心气阴两虚证心力衰竭大鼠血浆代谢组学特征。方法 将Dahl盐敏感性大鼠分为模型组和正常组,通过高盐饲料喂养模型组大鼠的方法制造心衰模型,于成模后采集模型组(7只)和正常组(8只)大鼠的血浆样本,运用气相色谱-质谱(GC-MS)联用技术及多元统计分析进行模式识别和差异代谢物的筛选。结果 主成分分析和偏最小二乘判别分析后得到的得分图显示两组血浆样本的代谢成分积分点完全分离,表明两组大鼠的代谢模式存在明显差异,正交-偏最小二乘判别分析发现,与正常组比较,模型组苏糖酸、半胱氨酸、鸟氨酸、丙氨酸、4-羟基-脯氨酸、苏氨酸、甲基半胱氨酸、天冬酰胺、花生四烯酸、胆固醇、苏糖醇、丙酮酸、焦谷氨酸浓度上升;色氨酸、亮氨酸、木糖醇、核糖醇、丙二酸、尿素浓度下降。结论 心气阴两虚证心衰大鼠模型血浆代谢组学出现变化,差异代谢物与氨基酸代谢、脂代谢、糖代谢紊乱和肝功能异常相关。

    〔Abstract〕 Objective To investigate the features of plasma metabonomics in rats with heart failure with heart-Qi Yin deficiency syndrome. Methods Dahl salt-sensitive rats were divided into model group and normal group. The rats in the model group were given high-salt fodder to establish a model of heart failure. After modeling, plasma samples were collected from 7 rats in the model group and 8 in the normal group, and gas chromatography-mass spectroscopy and a multivariate statistical analysis were performed for analysis. Results The score chart obtained by principal component analysis and partial least squares-discriminant analysis showed that the integral point of metabolic components was completely separated between the two groups, indicating that there was a significant difference in metabolic pattern between the two groups. The orthogonal partial least squares-discriminant analysis of the variables with between-group differences showed that compared with the normal group, the model group had significant increases in the concentrations of threonic acid, cysteine, ornithine, alanine, 4-hydroxy-proline, threonine, methyl cysteine, asparagine, arachidonic acid, cholesterol, D-threitol, pyruvic acid, and pyroglutamic acid and significant reductions in the concentrations of tryptophan, leucine, xylitol, ribitol, malonic acid, and carbamide. Conclusion There are changes in the plasma metabonomics of rats with heart failure, and differential metabolites are associated with disorders of amino acid metabolism, lipid metabolism, and glucose metabolism and abnormal liver function.

    代謝组学是继基因组学和蛋白质组学后新兴的研究方法,通过现代分析技术(液相色谱-质谱、气相色谱-质谱、核磁共振等)定量测定生物体中的内源性代谢产物,结合模式识别等化学信息学技术考察生物体在不同状态下代谢产物的变化,从而寻找代谢物与病理生理之间的联系[1],其具有整体性和系统性的特点,与中医学的整体观念相契合,因此可作为中医证候研究的出发点和切入点[2]。本研究运用基于气相色谱-质谱(GC-MS)联用的代谢组学技术对心气阴两虚证心衰大鼠的血浆代谢物进行研究,探讨其代谢组学变化规律,进一步揭示证候的生物学物质基础,为心衰的诊断、辨证和治疗提供新思路。
