急性应激对大鼠胸腺指数和血清CORT、胸腺肽浓度的影响(1)

     [摘要] 目的 研究急性应激对大鼠胸腺指数和血清糖皮质激素(CORT)和胸腺肽浓度的影响。方法 大鼠随机分为对照组和应激组。应激组大鼠给予急性束缚应激6 h。应激结束后取材,计算胸腺指数,酶联免疫吸附测定法检测血清CORT和胸腺肽的含量。 结果 应激组大鼠胸腺指数为(1.99±0.15)mg/g,与对照组(2.06±0.24)mg/g比较差异有统计学意义(P<0.05);应激组大鼠血清CORT浓度为(89.05±4.30)pg/mL,与对照组(67.82±4.76)pg/mL比较,差异有统计学意义(P<0.05);应激组大鼠血清胸腺肽的含量为(0.66±0.06)×103pg/mL,与对照组(1.46±0.08)×103pg/mL比较差异有统计学意义(P<0.05)。结论 急性应激会降低大鼠胸腺指数,抑制胸腺肽的分泌功能。

    Effects of Acute Stress on Thymus Index and Concentration of Serum CORT and Thymopeptide in Rats

    [Abstract] Objective To study the influence of acute stress on thymus index and concentration of serum CORT and thymopeptide in rats Methods Rats were randomly divided into control group and stress group. Acute restraint stress was imposed 6h in the stress group, and then the rats were killed and serum extracted for CORT and thymosin. We calculated the thymus index and determined the concentration of serum CORT and thymopeptide using enzyme-linked immunosorbent assay. Results The thymus index was (1.99±0.15)mg/g in the stress group and (2.06±0.24)mg/g in the control group, between which a statistically significant difference can be found, P﹤0.05; the CORT levels of stress group was(89.05±4.30)pg/ml in the stress group and(67.82±4.76)pg/ml in the control group, and the difference was statistically significant, P﹤0.05; the thymosin levels of serum was (0.66±0.06)×103pg/ml in the stress group and(1.46±0.08)×103pg/ml in the control group, and the difference was statistically significant, P﹤0.05. Conclusion Acute stress can affect the thymus index and inhibit the secretion of the thymosinin in rats.

    应激在我们的日常生活中非常普遍,几乎每一个人都会遇到某些应激源的作用。过强或持续时间比较长的应激对机体是有害的,影响机体的生理状态和免疫系统[1],应激对机体健康的影响越来越受到重视。机体在应激状态下,多出现以交感神经-肾上腺髓质轴和下丘脑-垂体-肾上腺皮质轴反应为主的变化,使血清肾上腺糖皮质激素含量增加,这常常被视为应激的标志[2-3]。

    胸腺是机体重要的中枢免疫器官,是T淋巴细胞发育、成熟的场所,对机体外周淋巴器官的发育和免疫应答的发生起着重要的调节作用[4]。应激反应可导致胸腺的退化萎缩[5],并伴有免疫功能受损。目前,对应激反应中血清胸腺肽浓度变化的研究较少。该研究试图通过急性应激探讨应激对大鼠胸腺指数和血清CORT以及胸腺肽浓度的影响,为进一步探讨应激影响机体免疫功能的机制与途径奠定基础。测定胸腺指数旨在了解应激对大鼠胸腺体积、重量的影响;检测应激后血清胸腺肽浓度,旨在了解应激对胸腺肽的影响,为进一步探讨胸腺肽在应激影响胸腺结构过程中的作用及其机制提供理论依据,现报道如下。, 百拇医药(崔春红 孙兆霞 李伟 姜鹤)
