HA作为药物载体合成HA-TGF-β1缓释剂对自体兔耳软骨移植影响的实验研究(1)

     [摘要] 目的 研究HA作为药物载体合成HA-TGF-β1缓释剂对软骨组织中软骨细胞增殖、Ⅱ型胶原的表达和GAG分泌含量的影响。 方法 将32只家兔随机分成2周组、4周组、8周组和12周组4个组别,每组8只。每只兔取耳软骨1 cm×1 cm 4块,分别植入自体背部4个皮下腔,按既定顺序给予腔内注射NS 2 mL、HA (100 μg/L)2 mL、TGF-β1(10 ng/mL)2 mL及TGF-β1(10 ng/mL)+ HA(100 μg/L)混合液2 mL,于移植后不同时期用HE染色观察软骨细胞的增殖与退变及边缘细胞的活力,Ⅱ型胶原免疫组织化学检测Ⅱ型胶原的表达及阿利新蓝比色法测定细胞外基质糖胺多糖(glycosaminoglycan, GAG)的含量。结果 在本实验条件下,软骨移植后,HA-TGF-β1组软骨细胞的增殖活跃,Ⅱ型胶原的表达呈强阳性,GAG的含量显著高于其他组(P < 0.05),差异有统计学意义。 结论 HA对TGF-β1有一定的缓释作用,使其缓慢释放,诱导软骨细胞的增殖,增加细胞的活力,从而使移植软骨更易成活。

    Experimental study of HA-TGF-β1 delayed release effect on the rabbits ear cartilage autograft

    [Abstract] Objective To study the influence of the multiplication of cartilage cells, the expression of collagen Ⅱ and GAG content on HA as a drug carrier to synthesise HA-TGF-β1. Methods In this investigation, 32 domestic rabbits were randomly divided into 4 groups: 2 weeks group, 4 weeks group, 8 weeks group and 12 weeks group, with 8 rabbits in each group. Every rabbit takes 4 pieces of its ear cartilage and those ear cartilages were implanted into back subcutaneous cavity. Then each habbit was injected NS 2 mL, HA ( 100 μg/L ) 2 mL, TGF-β1 (10 ng/mL) 2 mL and TGF-β1 (10 ng/mL) + HA (100 μg/L) mixture of 2 mL in the corresponding back subcutaneous cavity. In 2, 4, 8, 12 weeks,the multiplication, the degeneration and the edge cells viability of cartilage cells were observed with HE staining, the expression of collagen Ⅱwas determined by immunohistochemical detection, and glycosaminoglycan (GAG) content was measured by the alcian blue colorimetric method ......
