[摘要] 目的 比较蒸法和常规炮制方法对白芍中芍药苷含量的影响。 方法 采用RP-HPLC法测定两种方法制得的白芍中芍药苷的含量。色谱柱为waters symmetry C18(150×3.9 mm，5 μm)；以乙腈-0.1%磷酸水溶液(14∶86)为流动相；检测波长为230 nm；流速为1 mL/min。 结果 通过HPLC法测定蒸法和常规炮制法制得的三批芍药苷的含量分别为5.3%、5.7%、5.5%和3.9%、3.5%、3.4%。该方法的平均加样回收率为100.54%，其RSD为1.09%。并且精密度良好。 结论 白芍中芍药苷以蒸法提取较好，含量较高，方法简便快速。

The effect of HPLC determination of steaming method and conventional processing methods for the content of paeoniflorin in peony

[Abstract] Objective To compare the effect of HPLC determination of steaming method and conventional processing methods for the content of paeoniflorin in peony. Methods RP-HPLC method for the determination of the content of paeoniflorin in peony was taken. The column was the waters symmetry C18 (150×3.9 mm, 5 μm). Acetonitrile-0.1% phosphoric acid solution (14∶86) was taken as mobile phase. The wavelength was 230 nm by detection. The flow rate was 1 mL/min. Results The three batches of paeoniflorin by HPLC determination of evaporation and conventional processing system were 5.3%, 5.7%, 5.5% and 3.9%, 3.5%, 3.4%. The average recovery rate was 100.54%, and the RSD was 1.09%. And the precision was good. Conclusion Paeoniflorin in white peony extract has better evaporation, high content, and is simple and fast.

白芍在中国药典2010年版一部中收载，是毛茛科植物芍药(Paeunia lactiflora Pall.)的去干燥根[1]。性微寒，味苦、酸，归肝、脾经，具有养血调经、平肝止痛、敛阴止汗等多种活性[2]。芍药苷不仅是主要活性成分，并且也是中药饮片白芍质量标准的标定成分。加工方法不同可使白芍中的芍药苷含量变化显著[3]。本文以购自毫州的白芍中的芍药苷含量为指标，采用蒸法和常规炮制法进行比较，以RP-HPLC法进行测定，现将结果报道如下：
1 仪器与试剂
1.1 仪器
LC-20AD双泵高效液相色谱仪，SPD-M20A紫外检测器，SIL-20AC自动进样器，CTO-20A柱温箱，CBM-20A工作站，岛津AUW120D分析天平，岛津AUY220分析天平，3L机械定时超声波清洗器。
1.2 试剂
芍药苷对照品(批号：110736-200424，购自中国药品生物制品鉴定所)，白芍(购自亳州产白芍)，乙腈(色谱纯，Merck公司生产)，水为双重蒸馏水，其余试剂均为分析纯。
2 方法与结果
2.1 蒸法供试品制备
选择适量的原药材，剔除杂质，置于适宜容器内以蒸汽熏蒸30 min至软为宜，切片，干燥，粉碎，精密称量细粉0.1 g，置50 mL容量瓶中，加入稀乙醇35 mL，超声30 min，摇匀，放冷后，以0.45 μm微孔滤膜过滤，取续滤液，即得。同法制备3批样品 ......
