胆固醇酯转运蛋白基因单核苷酸多态与中国东北地区人群阿尔茨海默病的关系(2)
本研究发现:rs2303790AG对AD的减少作用主要表现在ApoEε4拥有者,而非ApoEε4拥有者则未见AD发生的减少,这又是什么原因呢?笔者推测可能是基因型AG弥补了ApoEε4所致的低HDL表达水平。其机制可能是:(1)ApoE基因与CEPT基因在HDL表达功能方面存在单纯的互补。ApoE的表达与其等位基因型关系密切,按表达HDL量的由多到少,ApoE等位基因的排序是:ApoEε2>ApoEε3>ApoEε4。很可能rs2303790AG通过弥补ApoEε4致脑内低HDL产出的缺陷,起到保护神经元的作用;(2)ApoE与CEPT在影响HDL脑内表达的过程中有着复杂的交互作用。有文献[7]证明:ApoE对HDL脑局部、脑脊液水平的影响与CEPT的单核苷酸多态有关,对rs708272的CC基因型来说:这种影响表现为ApoEε2>ApoEε3>ApoEε4;如果换成T碱基拥有者,却会呈现相反的结果:ApoEε2 , http://www.100md.com
关于CEPT的单核苷酸多态影响ApoE所致HDL产出的机制,目前尚不能阐明,可能的原因是ApoE对HDL表达水平的控制不仅与不同ApoE等位基因所引起的受体亲和效应的差异有关外[10-12],还可能与CEPT表达水平受不同ApoE等位基因的调控存在差别有关(ApoEε2>ApoEε3>ApoEε4),这两种方式同时存在。
总之,笔者的研究证实了CEPT基因rs2303790AG这种少见的基因型可以减少中国东北地区人群AD的发生,并且该作用在ApoEε4拥有者人群中更显著,这种作用是独立于年龄之外的独立性防止AD发生的保护作用。
