PA对人胃癌细胞放射敏感性的分析(1)
[摘要]目的 观察紫杉醇(PA)对人胃癌细胞株SGC7901放射敏感性的影响。方法 采用MTT法获得PA对SGC7901细胞的半数抑制浓度(IC50),并以20%的IC50剂量为增敏浓度;克隆形成实验检测单纯照射组与PA(增敏浓度)+照射组在0、2、4、6、8 Gy X线照射后的细胞存活率,并观察细胞周期的改变及细胞形态的变化。结果 PA对SGC7901细胞的IC50为16.11 μg/ml,随着照射剂量的逐渐增加,单纯照射组和PA+照射组细胞存活率呈下降趋势,从4 Gy开始,PA+照射组细胞的存活率低于单纯照射组(P<0.05);PA+照射组的G2/M期细胞比例高于其他两组,S期细胞比例低于其他两组(P<0.05)。结论 PA对胃癌细胞株SGC7901具有放射增敏作用,其机制可能与诱导细胞凋亡及G2/M期阻滞有关。
