抑癌基因PTEN的研究进展(1)
[摘要]抑癌基因PTEN具有双重特异性磷酸酶活性,其表达缺失可见于多种散发性肿瘤。PTEN基因主要通过对过磷脂酰肌醇3激酶(Pl3K)/蛋白激酶B(AKT)和丝裂原激活蛋白激酶(MAPK)等信号通路平衡的调节发挥抑癌机制。转录前(如基因突变、甲基化、表达缺失)以及转录水平和转录后的调控是PTEN基因表达的重要影响因素。本文就PTEN基因结构、生物学功能、与信号通路作用機制及其调控的研究进展作一综述。[关键词]PTEN;基因;肿瘤;进展
