放射线照射引起胶质瘤细胞细胞周期和凋亡的动力学变化及其对Chk1/2蛋白表达的影响(1)

【摘要】 目的:观察非同步化的U251细胞株对放射线所表现的细胞周期和凋亡的变化以及放射线对Chk1、Chk2蛋白表达的影响。方法:以不同剂量的放射线处理非同步化的U251细胞株,流式细胞仪检测处理后不同时间细胞周期和凋亡的变化,流式细胞仪和Western blot法检测照射后Chk1、Chk2蛋白表达量。结果:非同步化的U251细胞株经放射线处理后可引起G2/M期阻滞,细胞周期阻滞呈照射剂量依赖性增强,而细胞凋亡均发生于细胞周期阻滞解除后,其强度与细胞周期激活的程度成反比。照射后不影响Chk1、Chk2蛋白水平的表达。结论:放射线处理非同步化的U251细胞株可成功地建立G2/M期阻滞模型,但对Chk1、Chk2蛋白水平表达无影响。

【Abstract】 Objective: To observe the changes of cell cycle and the apoptosis of unsynchronized U251 cell line and effect on Chk1 and Chk2 protein after the irradiation. Methods: The changes of cell cycle and apoptosis in U251 cell line which were treated with irradiation after different intervals by flow cytometry were observed. The expression of Chk1 and Chk2 protein was measured by flow cytometry and Western blot. Results: G2/M cell arrest induced in unsynchronized tumor cells after treated with irradiation. Cell cycle arrest increased with the increased dosage of irradiation, and apoptosis happened after cell cycle arrest and had inverse relationship with the activating cell cycle. Irradiation had no effect on expression of Chk1 and Chk2 protein. Conclusion: G2/M cell arrest models successfully induced in unsynchronized U251 cell line after treated with irradiation.

放疗在胶质瘤临床治疗中占有举足轻重的地位,放疗的疗效好坏很大程度上取决于DNA损伤检测点的功能状态。目前国外研究多采用同步化处理的肿瘤细胞来建立G2/M阻滞模型[1],本研究采用更能真实反映细胞内在生物学特性的非同步化的肿瘤细胞,研究放疗作用下,胶质瘤细胞的细胞周期和凋亡的内在动力学变化并讨论放射线损伤对Chk1和Chk2蛋白表达的影响,以了解胶质瘤细胞中Chk1和Chk2对照射后细胞周期的调控作用。
1 材料与方法
1.1 细胞系培养及照射 本课题采用人胶质母细胞瘤U251细胞株,由武汉大学典型物保藏中心提供。在37 ℃恒温培养箱内对U251细胞进行复苏、培养至对数生长期,再进行传代。将5×105细胞接种于6孔板中,室温下采用瑞典Elekta公司SLi型直线加速器,250 Gy/min 6 MV X线照射分别给予6 、10 、15 Gy三种剂量的照射。于6、12、24、48、60 h收获细胞和上清,800 r/min离心5 min,加入75%冰乙醇1 ml固定,-20 ℃保存。
1.2 方法
1.2.1 流式细胞仪检测细胞周期及凋亡率(PI法) 取出固定的细胞,制成单细胞悬液,离心洗涤后加入75%的冰乙醇1 ml固定过夜 ......
