RMPP患儿肺泡灌洗液中巨噬细胞的表达及与IL-2、IL-8、IL-10的相关性(1)
【摘要】 目的:观察小儿难治性支原体肺炎(refractory mycoplasma pneumoniae pneumonia, RMPP)患儿支气管肺泡灌洗液(BALF)中巨噬细胞及IL-2、IL-8、IL-10水平的表达。方法:选取2014年10月-2018年10月在本院儿科住院治疗的94例RMPP患儿为RMPP组,选取同期轻型肺炎支原体肺炎(mycoplasma pneumoniae pneumonia, MPP)60例为MPP组及60例行支气管异物取出术儿童为对照组。ELISA检测BALF中IL-2、IL-8、IL-10水平,苏木精-伊红染色下计数巨噬细胞百分比。比较各组肺泡灌洗液中IL-2、IL-8、IL-10水平及巨噬细胞计数,分析RMPP患儿肺泡灌洗液中巨噬细胞计数与IL-2、IL-8、IL-10水平的相关性。结果:三组患儿BALF中IL-2、IL-8、IL-10含量比较,差异均有统计学意义(P<0.05)。RMPP組患儿BALF中IL-2、IL-8含量均高于对照组及MPP组,IL-10含量低于对照组及MPP组,差异均有统计学意义(P<0.05);MPP组患儿BALF中IL-2、IL-8含量均高于对照组,IL-10含量低于对照组,差异均有统计学意义(P<0.05)。MPP组患儿肺泡灌洗液中巨噬细胞数为(32.2±6.3)%,高于RMPP组的(27.6±5.7)%,低于对照组的(66.1±7.9)%,差异均有统计学意义(P<0.05);RMPP组患儿肺泡灌洗液中巨噬细胞数低于对照组,差异有统计学意义(P<0.05)。三组患儿肺泡灌洗液中巨噬细胞数比较,差异有统计学意义(P<0.05)。RMPP患儿肺泡灌洗液中巨噬细胞计数与IL-10水平呈正相关(P<0.05),与IL-2、IL-8水平均呈负相关(P<0.05)。结论:RMPP患儿BALF中的IL-2、IL-8含量升高,IL-10含量降低,与巨噬细胞百分比关系密切。

[Abstract] Objective: To observe the expression of macrophages and IL-2, IL-8 and IL-10 in bronchoalveolar lavage fluid (BALF) of children with refractory mycoplasma pneumoniae pneumonia (RMPP). Method: A total of 94 patients with RMPP hospitalized in our hospital from October 2014 to October 2018 were selected as the RMPP group. Sixty cases of mycoplasma pneumoniae pneumonia (MPP) in the same period were selected as MPP group and 60 children undergoing bronchial foreign body removal were selected as the control group. The levels of IL-2, IL-8 and IL-10 in BALF were detected by ELISA. The percentage of macrophages was counted by hematoxylin eosin staining. The levels of IL-2, IL-8, IL-10 and macrophage count in BALF of each group were compared, and the correlation between macrophage count and IL-2, IL-8 and IL-10 levels were analyzed. Result: There were significant differences in the contents of IL-2, IL-8 and IL-10 in BALF among the three groups (P<0.05). The contents of IL-2 and IL-8 in BALF of RMPP group were higher than those of control group and MPP group, and the content of IL-10 was lower than those of control group and MPP group, the differences were statistically significant (P<0.05). The contents of IL-2 and IL-8 in BALF of MPP group were higher than those of control group, and the content of IL-10 was lower than that of control group, the differences were statistically significant (P<0.05). The number of macrophages in BALF of MPP group was (32.2±6.3)%, which was higher than that of RMPP group (27.6±5.7)% and lower than that of control group (66.1±7.9)%, the differences were statistically significant (P<0.05). The number of macrophages in BALF of RMPP group was lower than that of control group, the difference was statistically significant (P<0.05). There were significant differences in the number of macrophages in BALF among the three groups (P<0.05). The number of macrophages in BALF was positively correlated with IL-10 (P<0.05), and negatively correlated with IL-2 and IL-8 (P<0.05).
