mTOR信号通路在皮肤恶性肿瘤中的研究进展

    【摘要】 哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)是一种丝氨酸/苏氨酸蛋白激酶,在细胞生长、增殖、分化、细胞周期调控等多个方面起到重要作用。近年来研究发现mTOR在皮肤恶性肿瘤(基底细胞癌、鳞状细胞癌、黑色素瘤、皮肤淋巴瘤、隆突性皮肤纤维肉瘤、血管肉瘤等)中异常表达,并通过影响肿瘤细胞增殖、凋亡、侵袭转移和血管生成等,广泛参与肿瘤的发生、进展和转移过程。mTOR的异常表达有望为皮肤恶性肿瘤的诊断和治疗提供新的方法。

    [Abstract] Mammalian target of rapamycin (mTOR) is a serine/threonine protein kinase that is centrally involved in the control of cell growth proliferation differentiation and cell cycle regulation. Recent studies indicate that mTOR is aberrantly expressed in cutaneous malignant tumors (basal cell carcinoma, squamous cell carcinoma, melanoma, cutaneous lymphoma, dermatofibrosarcoma protuberant, angiosarcoma, etc.), which is associated with cell proliferation, apoptosis, invasion, metastasis, and angiogenesis. mTOR is widely participated in the regulating tumor initiation, progression and metastasis, and its abnormal expression is expected to provide a new method for diagnosis and therapy of cutaneous malignant tumors.
