【摘要】目的研究CCK-8法检测兔筋膜成纤维细胞(RFFBs)增殖活性的最佳测试条件。方法以兔筋膜作为成纤维细胞原代培养组织,利用传代至P3的成纤维细胞,对比CCK-8法在不同孵育时间及不同检测波长条件下吸光光度值的差异,研究CCK-8法检测兔筋膜成纤维细胞增殖活性的最佳检测时间及最佳检测波长。结果CCK-8法检测兔筋膜成纤维细胞增殖活性的最佳检测时间为加入CCK-8试剂孵育后的4 h,最佳检测波长为450 nm。结论CCK-8法在孵育4 h后及酶标仪450 nm波长下检测兔筋膜成纤维细胞的灵敏度最高。
