高效液相色谱法测定人血浆中帕珠沙星浓度

【摘要】目的建立HPLC-紫外法检测人血浆中帕珠沙星浓度的方法。方法采用25%高氯酸溶液沉淀蛋白,分析柱为CNWSILC18(4.6 mm×100 mm),流动相为0.02 mol/L磷酸二氢钠溶液-乙腈-三乙胺(86∶13.5∶0.5),pH=3.5,紫外测定波长248 nm。结果帕珠沙星浓度在0.08~30.96 mg/L呈线性关系,r=0.9998(n=6);萃取回收率高于88%,方法回收率98%~101%,日间日内精密度均小于6%。在-40 ℃冰箱保存15 d或冻融2次稳定性较好。结论本方法简便灵敏,适合用来检测血浆中帕珠沙星浓度。
