肝癌组织中Sufu\Hip及Cyclin D1的表达及意义(2)
Cyclin D1由染色体11q13上的CCND1基因编码,含295个氨基酸,是高度保守的细胞周期蛋白家族成员之一。它调控细胞的有丝分裂,是G1期细胞周期蛋白。Cyclin D1基因的突变、扩增或过表达都将改变细胞周期的进程并可能导致肿瘤的形成,而且它还是多个信号通路下游的靶基因。在多种肿瘤中观察到Cyclin D1扩增和表达增强。众多研究提示肿瘤中Cyclin D1过表达则预后不良,Cyclin D1阳性患者的平均生存时间明显短于Cyclin D1阴性患者。有学者用Cyclin D1反义基因转染人类肿瘤细胞或利用RNA干扰可抑制肿瘤细胞增殖,降低其致瘤性。我们所检测的Sufu和Hip在肝癌组织中低表达与Cyclin D1表达增高二者之间并不具有相关性,说明Sufu及Hip表达异常可导致Hedgehog信号通路激活,但并不一定导致肝癌肿瘤细胞增值,亦说明肝癌肿瘤细胞的增殖不全由Hedgehog信号通路激活所致。
