膝关节炎治疗仪对SNP诱导体外大鼠软骨细胞凋亡的影响(1)
【摘 要】目的:观察膝关节炎治疗仪对硝普钠(SNP)誘导体外大鼠软骨细胞凋亡的影响。方法:采用膝关节炎治疗仪不同刺激强度干预1 mM SNP诱导第2代大鼠软骨细胞的凋亡模型,镜下观察软骨细胞形态,用噻唑蓝(MTT)法检测软骨细胞的活力,倒置荧光显微镜观察细胞核形态及密度变化。结果:倒置相差显微镜下观察膝关节炎治疗仪能减少细胞凋亡,MTT发现4级干预强度抑制细胞凋亡效果最佳,4,6-二脒基-2-苯基吲哚(DAPI)染色观察其能有效减轻软骨细胞核固缩。结论:膝关节炎治疗仪可能通过抑制软骨细胞凋亡从而达到防治骨关节炎的作用,但其具体抗凋亡途径有待进一步揭示。
